Useful For
PML/RARA fusion oncogene is present in >99% of cases of acute promyelocytric leukemia (APL). This real-time polymerase chain reaction assay can be used to detect the presence of the two gene fusion patterns: short and long isoform. It can be used to monitor minimal residual disease in APL. Ideally, recent patient samples should be tested side by side with the previous sample to determine if there is a change in disease course.
Methodology
Real-Time Reverse Transcriptase Polymerase Chain Reaction (RT-PCR)
Specimen Requirements
- Specimen must arrive within 72 hours of draw.
- Do not transport on weekends.
Submit only 1 of the following specimens:
Whole Blood
- Draw blood in an ACD solution A (yellow top) tube or sodium heparin (green top) tube
- Send 6 mL of unprocessed whole blood.
- Forward unprocessed whole blood promptly at ambient temperature only.
- Indicate date and time of draw and blood on request form.
- Label specimen appropriately (blood).
Bone Marrow
- Place 2 mL of bone marrow in a lavender-top (EDTA) tube(s)
- Forward promptly at ambient temperature only.
- Indicate date and time of collection and bone marrow on request form.
- Label specimen appropriately (bone marrow).
Reject Due To
- Clotted specimens (may not always be rejected)
- Hemolyzed specimens
- Frozen specimens
- Specimens > 72 hours old
